ABSTRACT

ObjectiveTo explore the effect and mechanism of Sishenwan-containing serum on aerobic glycolysis in human colon cancer HCT116 cells. MethodCell counting kit-8 (CCK-8) was used to detect the cell viability of colon cancer HCT116 cells after treatment with Sishenwan-containing serum (2.5%, 5%, and 10%) for 24, 48, 72 h. The concentration of lactic acid, the content of intracellular glucose, and the activity of hexokinase (HK) and fructose-6-phosphate kinase (PFK) in the cell culture medium were detected by the micro-method. The content of glucose transporter 1 (GluT1) mRNA was detected by Real-time quantitative polymerase chain reaction (Real-time PCR). The protein expression of GluT1 and methyltransferase-like 3 (MettL3) was detected by Western blot. The expression of GluT1 in cells was detected by immunofluorescence and the level of N6-methyladenosine (m6A) RNA methylation was detected by colorimetry. ResultCompared with the normal serum, 2.5%, 5%, and 10% Sishenwan-containing serum had no significant effect on the viability of HCT116 cells at 24 h, while 10% Sishenwan-containing serum showed a significant inhibitory effect on the viability of HCT116 cells at 48 h (P<0.05). Hence, 10% Sishenwan-containing serum was used in subsequent experiments, and the intervention time was 48 h. Compared with the normal serum, 10% Sishenwan-containing serum could reduce lactate production (P<0.05), down-regulate glucose uptake (P<0.05), and blunt the activities of HK and PFK, the key rate-limiting enzymes of glycolysis (P<0.05). Meanwhile, 10% Sishenwan-containing serum could decrease the expression of GluT1 protein (P<0.01) and mRNA (P<0.05) and reduce the proportion of cells expressing GluT1 (P<0.01). Compared with the normal serum, Sishenwan-containing serum also decreased the protein content of MettL3 (P<0.05) and the methylation level of m6A RNA (P<0.01). ConclusionSishenwan can inhibit glycolysis in colon cancer cells, and its inhibitory mechanism may be related to reducing MettL3 overexpression, inhibiting m6A RNA methylation, and down-regulating GluT1 and the activities of intracellular aerobic glycolysis-related enzymes such as HK and PFK.

ABSTRACT

Objective:To study the efficacy, safety and prognostic factors affecting survival of proton radiation therapy for primary hepatocellular carcinoma (HCC).Methods:Forty-four patients with primary hepatocellular carcinoma treated at Zibo Wanjie Cancer Hospital in Shandong Province from June 15, 2005 to September 12, 2018 were enrolled and given proton radiation therapy with a single dose of 2-5 Gy and a total dose of 25-75 Gy. The overall and local control rates, factors influencing survival, and adverse effects were observed during a follow-up period of 12 to 120 months.Results:The overall survival rates of all patients were 79.5% at 1 year, 68.2% at 2 years, 50.0% at 3 years, and 45.5% at 5 years. The overall local control rate was 95.5%. Patients older than 50 years had a longer overall survival (Log Rank Chi-Square of 4.787, P = 0.029). Child-Pugh A classification had better overall survival compared with B classification (Log Rank Chi-Square 4.077, P = 0.043). The main adverse reactions were skin reactions and gastrointestinal reactions. Among the patients with adverse reactions, 23 patients had skin reactions and 9 patients had mild gastrointestinal reactions, and no serious adverse events were found. Conclusions:Proton radiation therapy has good efficacy and high safety in patients with HCC. Age and Child-Pugh classification are prognostic factors affecting survival after proton radiation therapy.

ABSTRACT

Objective:To explore the effects of non-suicidal self-injury on suicidal ideation in adolescents with depressive disorder, and the mediating role of rumination and depression between them.Methods:A sample of 397 depressive disorder adolescents were recruited to complete the adolescent non-suicidal self-injury behaviour questionnaire, ruminative responses scale (RRS), self-rating depression scale (SDS), and Beck scale for suicide ideation-Chinese version(BSI-CV). All data processing and analysis were performed using SPSS 23.0.The mediating effect was tested by correlation analysis and Bootstrap analysis.Results:The non-suicidal self-injury score was (29.192±11.281), the rumination score was (65.036±12.284), the depression score was (75.770±11.278), and the suicidal ideation score was (40.681±11.626). Non-suicidal self-injury was significantly and positively correlated with suicidal ideation( r=0.403, P<0.01), rumination and depression( r=0.332, 0.470, both P<0.01). Rumination was significantly and positively correlated with depression and suicidal ideation( r=0.549, 0.181, both P<0.05). Depression was significantly and positively correlated with suicidal ideation( r=0.313, P<0.01). The direct effect value of non-suicidal self-injury on suicidal ideation was 0.341(95% CI=0.238-0.444), the indirect effect of non-suicidal self-injury on suicidal ideation through two pathways, the separate mediating effect value of depression was 0.057(95% CI=0.077-0.114), and the chain mediating effect value of rumination and depression was 0.026(95% CI=0.004-0.057). Conclusion:Non-suicidal self-injury can directly affect suicidal ideation of depressive disorder adolescents and indirectly through rumination and depression.

ABSTRACT

Objective:To analyze nine cases of pregnancy and lactation-associated osteoporosis(PLO) along with a literature review to improve clinicians′ knowledge of the disease.Methods:We collected medical history, laboratory examination, bone mineral density(BMD) scan, treatment and follow-up data of 9 inpatients with PLO in the Endocrinology Department of the First Affiliated Hospital of Zhengzhou University from January 2014 to June 2021.Results:The median age of onset of 9 patients with PLO was 30 years(22-37 years). All 9 patients presented low back pain, and the median time of low back pain after childbirth was 2.5 months(0.5-7 months). 89% of the patients presented vertebral compression fractures, and the mean number of vertebral fractures was 4.6. Eight patients with PLO showed osteoporosis by dual X-ray absorpiometry(DXA) scan, and one patient showed bone loss by quantitative CT scan. Osteoporosis predominated in the trabecular bone. After the diagnosis, nine patients with PLO stopped breastfeeding and were given calcium and vitamin D preparations. Seven patients were given bisphosphonates and one patient was treated with teriparatide followed by denosumab. The back pain of all patients was relieved, with the median of relief time being 3 months(7 d-6 m). After treatment initiation, BMD was increased in eight patients, especially obvious in the lumbar spine. No new clinical fractures occurred during the follow-up.Conclusions:For women with low back pain and shortened height in late pregnancy or breastfeeding, the possibility of PLO should be considered. Prompt diagnosis and early medical interventions are of utmost importance to reduce the risk of subsequent fractures and improve the prognosis.

ABSTRACT

Objective:To compare the predictive value of contrast-enhanced ultrasound(CEUS) Liver Imaging Reporting and Date System(LI-RADS) version 2017 with magnetic resonance imaging(MRI) LI-RADS version 2018 applied alone or in combination for focal liver lesions in high-risk patients.Methods:From January 2018 to October 2021, the clinical and imaging datas of 212 patients with 300 nodules underwent CEUS and contrast-enhanced MRI(CEMRI) within 4 weeks in Ruijin Hospital and its Wuxi branch were retrospectively analyzed. Each nodule was categorized according to the CEUS LI-RADS v2017 and CT/MRI LI-RADS v2018. Inter-modalities agreement was assessed with Cohen′s Kappa. The diagnostic performances of the two classification criteria applied alone and in combination for the predictive value of malignant risk of focal liver lesions were compared using histopathology or follow-up as gold standard.Results:The inter-modalities agreement of CEUS LI-RADS v2017 and MRI LI-RADS v2018 was moderate (Kappa=0.441). The specificity of CEUS LR-5, MRI LR-5 and CEUS LR-5+ MRI LR-5 in the diagnosis of HCC was 93.66%, 95.07% and 88.73% ( P>0.05), respectively, positive predictive values of them were 93.13%, 93.81% and 89.81%( P>0.05), respectively. The sensitivity of CEUS LR-M, MRI LR-M and CEUS LR-M+ MRI LR-M in the diagnosis of non-HCC malignancy was 85.71%, 82.86% and 100%, respectively. CEUS LR-M+ MRI LR-M had higher sensitivity than MRI LR-M( P=0.033), whereas no difference was found between CEUS LR-M+ MRI LR-M and CEUS LR-M( P=0.063). Conclusions:The inter-modalities agreement of the LI-RADS category between CEUS and MRI is moderate. The specificity and positive predictive values of HCC in LR-5 of the CEUS and MRI LI-RADS are comparable. In addition, the sensitivity of non-HCC malignancy in LR-M of the CEUS and MRI LI-RADS are comparable. The combined application of CEUS and MRI LR-M can improve the diagnostic sensitivity of non-HCC malignancy.

OBJECTIVE@#To detect potential variants of COL1A1 gene in five Chinese pedigrees affected with osteogenesis imperfecta (OI) and provide prenatal diagnosis for a fetus at 11th gestational week.@*METHODS@#The coding regions and exon/intron boundaries of 225 genes associated with bone diseases were subjected to targeted capture and next generation sequencing (NGS). Suspected mutations were verified with Sanger sequencing in the probands, unaffected relatives and 100 unrelated healthy controls. Prenatal diagnosis for a high-risk fetus was carried out by Sanger sequencing.@*RESULTS@#The probands of the pedigrees 1-5 have respectively carried c.3226G>A (p.Gly1076Ser), c.579delT (p.Gly194Valfs*71), c.2911-2912insAG (p.Gly971Glufs*138), c.3037G>A (p.Gly1013Arg) and c.642+5G>A variants of the COL1A1 gene. For pedigree 1, the same variant was not found in the fetus. c.3037G>A (p.Gly1013Arg) and c.2911-2912insAG (p.Gly971Glufs*138) were not reported previously.@*CONCLUSION@#Mutations of the COL1A1 gene probably underlie the OI in the five pedigrees. Combined NGS and Sanger sequencing can provide an effective and accurate method for the genetic and prenatal diagnosis of OI.

ABSTRACT

OBJECTIVE:To investigate the correlation between CYP2C19 gene polymorphisms and antiplatelet reactivity of clopidogrel in Xinjiang Uygur patients with acute coronary syndrome (ACS). METHODS :Totally 90 Uygur patients with ACS who were admitted to the cardiovascular department of Xinjiang Kashi Second People ’s Hospital from Jan. 2018 to Jan. 2019 were selected as the study subjects. They were given anti-platelet therapy of asprin+clopidogrel ,and received the treatment continuously for one year after discharge. The platelet aggregation inhibition (DPAI)rate of the patients were determined ,and the response to clopidogrel was evaluated. PCR-fluorescence probe method was used to detect genotype of CYP2C19*2 and * 3,and PCR-direct sequencing method was used to detect genotype of * 17. Multivariate Logistic regression analysis was adopted to investigate the correlation of gene and non-gene factors with DPAI of patients. RESULTS :Among 90 patients,there were 10 patients with clopidogrel resistance (CR)and 80 patients with non-CR. There were 58,28 and 4 patients with CYP2C19*2 G/G,G/A,A/A genotype,respectively;there were 88 and 2 patients with CYP2C19*3 G/G,G/A genotype ,respectively;there were 64 and 26 patients with CYP2C19*17 C/C,C/T genotype ,respectively;the genotype frequency of each genotype was consistent with Hardy-Weinberg equilibrium (P>0.05). After treatment ,DAPIs of patients with CYP2C19*2 G/A,A/A genotype were decreased significantly,while those of the patients with A/A genotype were significantly lower than patients with G/A genotype patients (P< 0.05). DAPIs of patients with CYP2C19*17 C/T genotype were increased significantly ,compared with C/C genotype 2016D01C087) patients (P<0.05). There was no statistical significance in @fudan.edu.cn DAPIs between CYP2C19*3 G/A and A/A genotype patients (P>0.05). Multivariate Logistic regression analysis showed 85775264@qq.com that CYP2C19 gene pol ymorphism was independently related to DPAI in Xinjiang Uygur patients with ACS [OR =2.314,95%CI(1.569,3.144),P=0.009],while age ,gender,smoking and other non-gene factors were not related to DPAI (P>0.05). CONCLUSIONS :CYP2C19 gene polymorphism is associated with antiplatelet reactivity of clopidogrel in Xinjiang Uygur patients with ACS. * 2 wild type patients may have higher DPAI ,while * 17 wild type patients may have lower DPAI.

ABSTRACT

Objective To investigate the effect of scan table on size-specific dose estimate ( size-specific dose estimate, SSDE) in children's CT scan. Methods CT imaging data and CTDIvol of 44 children ( 15 heads, 13 chests, 16 abdomen-pelvis) who underwent Siemens SOMATOM Definition AS+ 64 row 128-slice CT scan were retrospectively collected. CTDIvol of each patient was recored, WED ( water equivalent diameter) was calculated by two different methods ( with or without table) , donated as WED-T and WED-NT, then the corresponding SSDEWED ( SSDEWED-T and SSDEWED-NT ) was calculated. And the SSDEWED-NT was used as reference to evaluate the difference between WED and SSDEWED obtained by two different methods. Results Including part of table will lead to the overestimate for WED, with mean differences of 0. 10%, 2. 82% and 2. 54% for head, chest and abdomen-pelvis, respectively, while SSDEWED will be underestimated by 0. 06% ( head ) , 2. 70% ( chest ) and 1. 59% ( abdomen-pelvis ) . Conclusions Including par of the patient table has a certain effect on SSDEWED for children, more attention should be paid for the application of SSDEWED.

ABSTRACT

Objective To compare the renal toxicity of vancomycin with continuous infusion vs intermittent infusion. Methods The databases of EMBASE,PUBMED,the Cochrane Register of Controlled Trials,CBM,CNKI and WanFang were searched.The Cochrane Revman5.2 software was used for Meta-analysis.Results Two RCTs and eight observational studies were included in the systematic literature search with total of 1 764 patients.1 037 patients received vancomycin with continuous infusion while 727 patients with intermittent infusion.The Meta-analysis indicated that there was no significant difference in renal toxicity between continuous infusion group and intermittent infusion group(P>0.05).Conclusion Vanco-mycin continuous infusion cannot effectively reduce the incidence of renal toxicity.

ABSTRACT

Objective To explore the effect of a Chinese Medicine Xionggui Liujunzi Decoction on the experimental coronary heart disease in rats. Methods Rats were randomly divided into five groups: the blank control group, the model control group, the western medicine control group (simvastatin), the Chinese medicine control group ( compound Danshen dripping pills) and the Xionggui Liujunzi Decoction group. The coronary heart disease was induced by intragastric gavage of fat emulsion (10 mL/kg, q. d. for 12 weeks) and pituitrin (30 U/kg, q. d. for 3 days) was intraperitoneally injected to induce coronary artery spasm. Changes of the ST segment in electrocardiogram (ECG) and the blood lipids were detected, and the levels of the inflammatory factors including tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), interleukin-10 (IL-10), highly sensitive C-reactive protein (hs-CRP), monocyte chemotactic protein-1 (MCP-1) and intercellular adhesion molecule-1 (ICAM-1), the vascular endothelial active substances including ET-1, NO, TXA2 and PGI2, and the indicators of fibrinolytic system function such as PAI-1, t-PA in the plasma were measured. Results Compared with the model control group, each drug treatment showed better effects on the ST segment of the electrocardiogram, blood lipids, levels of the inflammatory factors and vascular endothelial active substances, and the fibrinolytic function, with significant differences (P < 0. 05). In particular, Xionggui Liujunzi Decoction has a significantly better effect than the compound Danshen dripping pills and simvastatin (P< 0. 05). Conclusions Xionggui Liujunzi Decoction can improve the ST segment of the electrocardiogram, blood lipids, levels of the vascular endothelial active substances and fibrinolytic function of the rat model of coronary heart disease, and alleviate inflammation responses, showing a significant effect on coronary arteriosclerosis and myocardial ischemia in rats.

ABSTRACT

Radioembolization with yttrium-90(90Y) microspheres is an alternative treatment choice for hepatic malignancies and has become incorporated into guidelines by many international oncology and other professional organizations. The tumoricidal effects of radioembolization are predominantly derived from its radioactivity rather than ischemia. Therefore, the overall side effects that have been associated with this therapy, such as fatigue, nausea and vomiting, which are mild but common. Occasionally, severe complications may occur due to incidental embolization of non-target tissue by the microspheres. This review describes in detail the manifestation of side effects and various complications that may be associated with radioembolization using 90Y microspheres. Management and treatment options of these potential adverse events will also be discussed briefly.

Accurate doses of yttrium-90 (90Y) microspheres are critical for the treatment of liver malignancies,because it is closely related to the clinical efficacy and adverse reactions.The dose calculationis primarily based on the type of ~Y delivery medium,whether it is glass microsphere (TheraSpheres) or resin microsphere (SIR-Spheres).The dose calculation of glass microspheres is based on the assumption that the expected radiation dose of the liver and the microspheres can be evenly distributed throughout the liver,while the dose calculation of resin microspheres is based on the assumption that the microspheres are not uniformly distributed throughout the liver,and the degree of this inhomogeneous distribution depends on the extent to which the normal liver is replaced by the tumor tissue.Many other factors may also potentially affect the therapeutic dose of 90Y microspheres.This review will introduce the calculation methods of 90Y microsphere dose,and describe the factors that need to be considered in order to achieve maximum efficacy and avoid adverse effects.

ABSTRACT

Objective Rats were fed with high-fat diet and were successfully induced the models with non-alcoholic fatty liver disease,and to investigate the effects of liraglutide on the expression of ERp46.Methods Thirty male Sprague-Dawley rats were assigned to normal chow group(NC,n=10)and high-fat diet group(n=20),after 12 weeks,the high-fat diet group rats were divided into high-fat diet group(HF,n=10)and liraglutide group(100L,n=10) and treated with normal saline and liraglutide(100 μg/kg)for 4 weeks respectively. Liver tissues were measured by hematoxylin-eosin(HE),Oil Red O staining,hepatic triglyceride(TG),and hyperinsulinemic-euglycemic clamp test(HECT). Hepatocyte apoptosis rate were evaluated by TdT-mediated dUTP nick-end labeling(TUNEL),and the expressions of ERp46 mRNA and protein were measured.Results Compared with the NC group,the liver tissues in the HF group have steatosis,insulin resistance,and the percentage of apoptosis was significantly increased. ERp46 mRNA and protein expressions were decreased(P<0.05). Compared with the HF group,liraglutide treatment was sufficient to reduce steatosis,insulin resistance,apoptosis,and increase the ERp46 mRNA and protein(P<0.05). Furthermore,the expression of ERp46 protein in the liver was negatively correlated with hepatocyte apoptosis rate,and positive correlated with glucose infusion rate(GIR, P<0. 05). Conclusion Liraglutide may up-regulate the expression of ERp46 to improve IR and hepatocyte apoptosis in NAFLD rats.

Through castrated rats osteoporosis models, this study aims to explore whether the intraperitoneal injection of Scutellaria baicalensis extract could prevent the occurrence of osteoporosis and slow down/reverse osteoporosis or not, and further explore the influence on BMMSCs extracted by Scutellaria baicalensis extract. Scutellaria baicalensis extracts on the prevention and treatment of oestrogen shortage and osteoporosis. After the screening of appropriate concentration, in the model of castrated female rats with osteoporosis, rats in prevent group were given intraperitoneal injection of Scutellaria baicalensis extract within 1 week after the castration, and the rats in treatment group were injected Scutellaria baicalensis extract at the abdominal cavity in two months of castration. It required 2 times per week, 12 weeks in total, Micro CT scanning was performed respectively at 6[th] and 12[th] week on one side of the femur to compare the differences in bone mass and bone trabecular quantity, in order to evaluate the prevention and treatment of Scutellaria baicalensis extract in osteoporosis. After intraperitoneal injection of Scutellaria baicalensis extract, Micro CT results showed the animal bone mass and bone trabecular quantity in prevention and treatment group were increased compared to control group, while, it only reduced bone loss and restored bone mass to a certain extent. In this study, the small molecular compound with higher content of flavonoids through leaching, extraction, enrichment method from Traditional Chinese medicine radix scutellariae and could be utilized on the prevention and treatment of osteoporosis caused by rats castration

ABSTRACT

Radioembolization with yttrium-90 (90Y) microspheres is a locoregional procedure during which either resin or glass microspheres loaded with β-emitting 90Y are selectively injected into a hepatic artery.The microspheres are trapped in downstream vasculatures,at which point they deliver a cytotoxic dose of radiation to the target tumor.The safety and efficacy of this method have been demonstrated in studies of survival outcomes in patients with nonresectable hepatic malignancies.In this first part of a series of reviews on radioembolization with 90Y microspheres,we cover the basic characteristics of 90Y and 90Y microspheres,techniques for the radioembolization procedure,and methods for ensuring radiation safety and protection.

ABSTRACT

Objective To explore the effects of mindful- based stress reduction exercise on the sleep and negative emotion in Primary glaucoma patients. Methods By using random sampling,primary glaucoma patients at the hospital of between September in 2015 and September in 2016. A total of 93 patients were divided into intervention group and control group according to random number table.The control group was given conventional nursing, intervention group joined in mindful-based stress reduction exercise on the basis of routine nursing. Baseline levels of anxiety, depression and status of sleep were measured in the two groups after admission, evaluated and comparison at the end of formal intervention. Results At the end of intervention, the score of Hamilton Anxiety Scale (HAMA) in intervention group was decreased from (13.42 ± 3.56) points to (8.23 ± 2.11) points (t=7.832, P=0.000), the score of Hamilton Depression Scale (HAMD) was decreased from (18.42 ± 4.55) points to (10.31 ± 2.76) points (t=9.517, P=0.000), the effect was better than the control group (t=8.117, 7.671, all P=0.000). And the score of Self-Rating Scale of Sleep (SRSS) was decreased from (30.81±8.35) points to(25.52±6.13) points (t=3.189, P=0.002), the effect was better than the control group (t=2.253, P=0.027). Conclusions Mindful-based stress reduction exercise can reduce negative emotions such as anxiety and depression and promote the status of sleep effectively.

Objective To investigate the effects of Qibai Pingfei Capsules(QPC) on the expression of monoamine neurotransmitters of 5-hydroxytryptamine receptor(5-HT) and dopamine(DA) receptors of DA1 and DA5 in the rats with chronic obstructive pulmonary disease (COPD) accompanied with depression, and to explore the possible therapeutic mechanism. Methods Ninety male Wistar rats were randomly divided into 6 groups, namely normal group, COPD depression group, QPC group, Venlafaxine group, and QPC plus Venlafaxine group. The rat model of COPD accompanied with depression was established by compound methods of smoking, food deprivation, water deprivation, forced swimming, shaking, and clipping tail. And then the effects of QPC on the lung function and the general characteristics were observed. Furthermore, the mRNA level of 5-HT1AR was determined by real-time quantitative reverse transcription PCR(RT-qPCR), and protein levels of DA-1 and DA-5 in rat hippocampus were assayed by Western blot analysis. Results Compared with the normal group, forced expiratory volume in 0.3 second(FEV0.3), the forced volume capacity(FVC) and FEV0.3/ FVC in the COPD depression group were significantly reduced (P < 0.01). Compared with the COPD depression group, the values mentioned above were restored to different extents in the groups of QPC, Venlafaxine, and QPC plus Venlafaxine(P<0.01). Compared with the normal group, the expression levels of 5-HT1AR mRNA, and DA-1 and DA-5 protein in the COPD depression group were significantly down-regulated, and their levels were also slightly decreased in the treatment groups(P<0.05). Compared with the COPD depression group, the expression levels were up-regulated to various degrees in all of the treatment groups (P < 0.05). Conclusion QPC can increase the mRNA levels of 5-HT1AR and protein levels of DA-1 and DA-5 in the hippocampus of COPD with depression.

ABSTRACT

Potential complications arising from yttrium-90 (90Y) radioembolization are often related to inadvertent embolization of non-target vasculature during particle administration.Therefore,careful pretherapeutic planning with arterial mapping is especially important to help identify potential high-risk arteries and vascular communications.A complete pre-therapeutic evaluation of hepatic arterial vasculature includes selective arteriography,precautionary embolization of potential risk arteries and identification of occurrences of hepatopulmonary shunting secondary to tumor-related pathologic arteriovenous channels.The aim of this review is to discuss the pertinent arterial anatomy during 90Y radioembolization therapy and strategies on how to evaluate the risk and prevent the occurrence of non-target embolization through those vascular structures.

Yttrium-90 (90Y) radioembolization therapy can directly deliver a high dose of radiation to the tumor being targeted,which has been demonstrated to be effective in treating patients with hepatic malignancy.Compared with transarterial chemoembolization,90Yradioembolization is less frequently associated with toxicities such as abdominal pain,fever,nausea,and vomiting.However,the efficacy and safety of 90Y radioembolization therapy are largely reliant on careful preprocedural evaluation and diligent follow-up.This review discusses the details of essential preprocedural work-up and postprocedural follow-up for patients treated with 90Y radioembolization.

ABSTRACT

Objective To observe the effect of TanshinoneⅡA( TanⅡA) on endometriosis( EMS) rat model and study the possible mechanism. Methods EMS rat model was established by autologous endometrium transplantation. EMS rats were randomly divided into five groups: high-, medium-, low-dose TanⅡA group, mifepristone group and model control group( n=10), the other ten normal rats as normal control group.The rats in the model control group and normal control group were given 0.9% sodium chloride solution. The rats in the mifepristone group were given 25 mg . kg-1 . d-1 mifepristone. The rats in the high-, medium-, low-dose TanⅡA groups were given 30, 20 and 10 mg.kg-1.d-1 TanⅡA, respectively.Four weeks after the treatment, the volume of endometriotic implants in each rat was measured. Pathological changes of ectopic endometrium were observed under light microscope.Expression levels of Bcl-2, Bax and Caspase-9 in ectopic endomembrane of rats were determined by Western blotting. Results High and medium dose TanⅡA and mifepristone significantly reduced the volume of ectopic lesions(P<0.05).As compared with model control group, the inhibition rate of the ectopic lesions in low-, medium-, high-dose TanⅡA groups and mifepristone group was 17.81%, 57.78%, 58.88% and 64.12%, respectively.High and medium dose TanⅡA and mifepristone promoted atrophy of ectopic endometrium, inhibited the expression of Bcl-2 protein and promoted the expression of Bax and Caspase-9 in ectopic tissues( P<0.05) . Conclusion TanⅡA significantly decreases the size of ectopic lesions in rats, possibly through reducing the expression of Bcl-2 and increasing the expression of Bax and Caspase-9 to regulate cell apoptosis.
